Research
여섯 가지 주제를 연구합니다. 약물을 연결하는 방법과 약물이 활성화되는 조건을 설계하고, 세포와 동물 모델에서 효능과 면역 반응을 평가합니다.Six research topics: how the drug is attached and under what condition it becomes active, evaluated for efficacy and immune response in cells and animal models.
01 · 펩타이드–약물 접합체와 자가분해 링커Peptide–drug conjugates and self-immolative linkers
펩타이드와 약물을 링커로 연결하고, 특정 효소나 자극이 있을 때만 약물이 활성화되도록 설계합니다.A peptide and a drug are joined by a linker, designed so that the drug is released only in the presence of a specific enzyme or stimulus.
일반적인 항암제는 전신에 분포하기 때문에 종양뿐 아니라 정상 조직에서도 독성을 나타냅니다.Conventional anticancer agents distribute through the whole body, so they are toxic to normal tissue as well as to the tumour.
주로 자가분해 링커(self-immolative linker)를 사용합니다. 절단 신호를 받으면 링커가 연쇄적으로 분해되면서 약물이 떨어져 나옵니다.The main tool is the self-immolative linker. Once the cleavage signal arrives, the linker decomposes in a cascade and the drug is released.
절단 신호는 기질 서열 설계로 바꿀 수 있습니다. 종양에서 발현되는 효소, 면역세포가 분비하는 효소, 외부에서 조사하는 방사선 등을 활용합니다.The cleavage signal can be changed by substrate sequence design: enzymes expressed in tumours, enzymes secreted by immune cells, or externally applied radiation.
02 · 항체–약물 접합체와 링커 설계Antibody–drug conjugates and linker design
항체의 어느 위치에 약물을 결합할지, 어떤 링커를 사용할지를 설계합니다. 한 항체에 약물을 더 많이 붙이면서도 응집이 일어나지 않는 링커를 개발합니다.We design where on an antibody the drug is attached and which linker joins them, aiming for linkers that raise the payload without causing aggregation.
대부분의 약물은 소수성이 강합니다. 한 항체에 여러 개를 결합하면 서로 응집해 사용할 수 없게 되므로, 지금 임상에서 쓰이는 접합체는 한 항체에 붙일 수 있는 약물 수가 제한적입니다.Most payloads are strongly hydrophobic. Attaching several to one antibody causes aggregation, which is why the payload of conjugates now in clinical use is limited.
전하를 띠는 친수성 펩타이드를 링커로 사용합니다. 링커의 수화층이 약물 간 응집을 억제하므로, 같은 항체에 더 많은 약물을 결합하고도 용해도를 유지할 수 있습니다.We use a charged hydrophilic peptide as the linker. Its hydration layer suppresses aggregation between payloads, so more drug can be carried on the same antibody while solubility is retained.
결합 위치도 함께 설계합니다. 부위특이적으로 결합하면 접합체의 균일도가 높아져 효능과 안전성을 예측하기 쉬워집니다.The attachment site is designed as well. Site-specific conjugation gives a more uniform product, which makes potency and safety easier to predict.
03 · Switchable 단백질Switchable proteins
기존 단백질의 결합 부위를 화학적으로 차단해 두었다가, 특정 신호가 있을 때 활성이 회복되도록 만듭니다.The binding site of an existing protein is blocked chemically, and the activity is restored when a specific signal appears.
항체는 투여 후 전신에서 활성을 유지하기 때문에 병변과 정상 조직을 구분하지 못합니다.An antibody stays active throughout the body after dosing, so it cannot distinguish a lesion from healthy tissue.
단백질을 새로 발현하지 않고, 이미 확보한 단백질에 화학적 변형을 가합니다. 결합 부위를 가리는 차단기를 절단형 링커로 연결해, 병변에서 링커가 끊어지면 차단기가 떨어지도록 설계합니다.Rather than expressing a new protein, we apply chemical modification to one already in hand: a blocking group covers the binding site through a cleavable linker, and the block falls away when the linker is cut at the lesion.
단백질 공학보다는 접합 화학에 가깝습니다. 차단 부위, 링커 구조, 활성화 신호를 설계 변수로 다룹니다.This is closer to conjugation chemistry than to protein engineering: the blocked site, the linker structure and the activating signal are the design variables.
04 · 바이오소재–펩타이드 접합체Biomaterial–peptide conjugates
펩타이드를 고분자·나노소재에 결합하고, 표적 부위에서 자가조립이 일어나도록 설계합니다.Peptides are conjugated to polymers and nanomaterials and designed to self-assemble at the target site.
면역세포의 반응은 입자의 크기, 형태, 강성에 따라 달라지는 것으로 알려져 있습니다. 같은 성분이라도 어떤 구조로 제시되느냐에 따라 결과가 달라집니다.Immune cell responses are known to depend on the size, shape and stiffness of a particle. With the same components, the structure in which they are presented changes the outcome.
용액 상태에서는 분산되어 있다가 표적 부위에서 조립되는 분자를 설계하고, 생성된 구조가 면역 반응에 미치는 영향을 평가합니다.We design molecules that stay dispersed in solution and assemble at the target site, then evaluate how the resulting structure affects the immune response.
05 · 펩타이드 PROTACPeptide PROTACs
펩타이드로 표적 단백질에 결합한 뒤, 세포 내 분해 기구로 유도해 단백질을 제거합니다.A peptide binds the target protein and routes it to the cell's degradation machinery, removing the protein.
면역 조절에 관여하는 단백질 중 상당수는 소분자가 결합할 포켓이 없어 약물 개발이 어려웠습니다.Many proteins involved in immune regulation have no pocket for a small molecule to bind, which has made them hard to drug.
펩타이드는 소분자가 접근하기 어려운 넓은 상호작용 표면에 결합할 수 있습니다. 관건은 펩타이드가 세포 안까지 들어가는 것이며, 접합체 연구에서 쌓은 방법을 적용합니다.Peptides can bind the broad interaction surfaces that small molecules cannot reach. The challenge is getting the peptide inside the cell, where the know-how built up in conjugate work is applied.
06 · 방사성핵종 접합체Radionuclide conjugates
킬레이터에 방사성 핵종을 결합하고, 표적 펩타이드나 항체에 연결해 종양에 모이게 합니다.A radionuclide is held in a chelator and linked to a targeting peptide or antibody so that it collects in the tumour.
같은 분자로 영상과 치료가 모두 가능합니다. 진단용 핵종을 결합하면 종양의 위치와 분포를 확인할 수 있고, 치료용 핵종으로 교체하면 같은 부위를 치료할 수 있습니다.One molecule serves for both imaging and therapy: with a diagnostic nuclide the location and extent of the tumour can be seen, and with a therapeutic nuclide the same site can be treated.
킬레이터 설계와 접합 화학이 핵심입니다. 핵종이 유리되면 표적이 아닌 조직에 방사선이 축적되기 때문입니다. 유기화학과 무기핵약학이 만나는 분야입니다.Chelator design and conjugation chemistry are decisive, because a nuclide that comes loose deposits radiation in the wrong tissue. This is where organic chemistry meets nuclear pharmacy.
